Impacts of plant growth regulators and culture media on in vitro propagation of three apple (Malus domestica Borkh.) rootstocks

author

Abstract:

Apple is one of the most important temperate-zone fruit trees in the world. Micropropagation of apple scion and rootstocks is an important method of rapid production of healthy and disease-free plants. In this research, in vitro propagation of three apple roottocks including Azayesh-Esfahan, Morabbaee-Mashhad and M9 was investigated. Shoot proliferation carried out in two basal media (MS and WPM) containing three concentrations of BA (0.5, 1 and 1.5 mgL-1) and also, rooting of mi-croshoots were investigated in two basal media (MS and ½ MS) with three concentrations of IBA (0.5, 1 and 1.5 mgL-1). The results showed that all studied factors including cultivars, media, BAP, IBA concentrations and interaction among them had significant effects on both shoot proliferation and rooting of rootstocks. Regardless of root-stocks and media, the maximum and minimum shoot proliferation rates were obtained in the me-dia containing 1.5 mgL-1 and 0.5 mgL-1 BA, re-spectively. The MS medium was more effective on shoot proliferation than WPM medium. The Azayesh-Esfahan and Morabbaee-Mashhad showed the maximum and minimum (4.46 and 3.66 shoots/explants) shoot proliferation values, respectively. However, all roottocks had the max-imum rooting in the ½MS media containing 1.5 mgL-1 IBA. In overall, Azayesh-Esfahan showed the maximum shoot proliferation (5.11 shoots/explants) and rooting (48.33 %) among roottocks.

Influence of Plant Growth Regulators and Media on In vitro Propagation of Sorbus aucuparia L.

Aucuparia is an important slow growth forest tree with medicinal, industrial and ornamental uses which is applied for reforestation in high altitude of mountain lands. This species has been endangered at northern forests of Iran; therefore micropropagation of adult trees by bud culture may help to reforestation. The best sterilizing treatment was the buds washing with HgCl2% 0.1 solution for 7 ...

Triterpenes and Phenolic Compounds in Apple Fruit (Malus domestica Borkh.)

Apple (Malus × domestica Borkh.), a popular and widely cultivated fruit world-wide, contains bioactive triterpenes and phenolic compounds with potentially valuable pharmacological functions. This thesis investigated the effects of pre-harvest and postharvest factors on concentrations of these bioactive compounds in apples. It also studied the effect of ozone treatment, before or during storage,...

Improvement of In vitro Proliferation of Apple (Malus domestica Borkh.) by Enriched Nano Chelated Iron Fertilizer

Nano-fertilizers can increase value of products in agriculture. Iron plays many important and essential roles in plant growth and development as compared to other micronutrients. In the present study, effects of different levels of enriched nano chelated Iron fertilizer (25, 50, 100, and 200 mg l-1)  were investigated in comparison with the common Iron (FeSO4.7H2O) on in vitro proliferation of ...

Effect of Antioxidants, Amino Acids and Plant Growth Regulators on in vitro Propagation of Rosa centifolia

Background: Rosa centifoliais commercially propagated by asexual means but in vitro propagation ensure the production of disease free and healthy plants and browning of explants creates hurdle in their multiplication. Objectives: The aim was to reduce oxidative browning of shoots of R. centifolia in MS medium during in vitro propagation. Materials and Methods: Axillary buds of R. centifolia we...

Effects of Apple (Malus domestica Borkh.) Diet on Rat Reproduction and Sex Ratio of Offsprings

Background & Aims: Apple (Malus domestica Borkh. from Rosaceae family), a rich source of phytoestrogenes has not been thoroughly tested for its reproductive effects. In this study, we investigated the effects of apple diet on rats' reproduction. Method: Nine groups of NMRI rats (n= 10 females + 2 males) with different apple diets (free- apple diet and apple diet in every other day and every tow...

Impact of shelf life on content of primary and secondary metabolites in apple (Malus domestica Borkh.).

In this study, we evaluated the changes in apple fruit quality during shelf life. After a month of cold storage, apples of cultivars "Jonagold" and "Golden Delicious" were exposed to ambient temperatures for 21 d, with subsequent sampling every 3 or 4 d. Fruit firmness, changes in amounts of sugars, malic acid, and phenolics were observed during shelf life. Chemical analyses were done with HPLC...
